Question:

We purchase your CMV pp28 antibody (5C3) via your US distributor. We have had a customer ask us what fixative was used on the CG015 fibroblast cell lines that were used to obtain the immunofluorescence image on the datasheet? He asks this because he is getting non-specific staining in his own fibroblast U373 cell lines and wondered if the problem was linked to the fixative that he's using (he's tried both PFA and methanol)? Also, at what dilution does your antibody give a 5+ signal? Many thanks for all of your help. It is very much appreciated.

Resolution:

Hi Simon,

The slides used in the testing of CMV antibodies are acetone fixed. The cells are CMV (AD169) infected human foreskin fibroblasts.

This antibody at 1 mg/ml gives a 4+ reaction at 1:200 and below when using our anti-mouse IgG-FITC (AG002) conjugate.

Hope this helps.
