Question:

  1. How much does it cost if we don't need additional eggs or extended maintenance?
  2. How much of antigen do we need to provide for immunization?
  3. Is any conjugation or modification necessary when using peptides as antigens?
  4. If so, what do we need to prepare?
  5. In terms of purity, is a peptide with >70% HPLC purity good enough?
  6. Is it possible to receive some eggs sooner than the end of 12-week period

Resolution:

I have addressed each of your questions below:

  1. How much does it cost if we don't need additional eggs or extended maintenance? Total price for the animal work is $575 for 2 hens - 3 pre and 6 hyperimmune eggs from each
  2. How much of antigen do we need to provide for immunization? Approximately 1 milligram total - we need 5 vials of 150 - 200 µg each
  3. Is any conjugation or modification necessary when using peptides as antigens? Yes - peptides are too small to elicit an immune response. Conjugation is to KLH and is monitored by a quantitative Ellmans assay.
  4. If so, what do we need to prepare? A peptide with a terminal reduced cysteine. We can make this for you @ $23/residue if you like
  5. In terms of purity, is a peptide with >70% HPLC purity good enough? 70% is fine for immunizations
  6. Is it possible to receive some eggs sooner than the end of 12-week period? The 10 - 12 week period includes the time needed for purification. We can get you hyperimmune eggs at roughly week 7 - 8.
