Rotavirus Inactivation


Question:


Do you have a protocol for virus inactivation. I am working with SA11 rotavirus.
I will appreciate all the help I can get.


Resolution:

Inactivation of virus infectivity depends upon many variables. There is not one universally applicable methodology.
Rotavirus infectivity is relatively stable to inactivation pH 3 - 9 and temperature to 50°C. Human strains seem to lose their outer capsid easier than bovine or simian strains.
We would work with SDS 0.1% or more. Do not use DOC or non-ionic detergents, as the former does not work and the latter can enhance infectivity.
Virus infectivity can be inactivated by disinfectants such as phenols, formalin, chlorine and betapropiolactone. Ethanol (95%) is perhaps the most effective, exerts its effect by removing the outer capsid layer.
