Client supplied recombinant baculovirus is plaque purified on a lawn of Sf-9 insect cells. The principle of the method is to infect cells with extremely low numbers of infectious virus particles so that only isolated cells become infected. The cells are overlaid with a solid medium to limit the spread of virus particles. After several cycles of infection, a focus of infection (plaque) is formed. All of the virus in this single plaque derives from a single particle and is therefore a clonal population. Sequential repetition of this isolation of clonal virus is the basis of the purification and is necessary to ensure removal of defective virions to produce an infectious stock.
